Hi all

i am trying to develop spheroids using Caco-2 cell line. I used Ultra low attachment plates from corning where i added 50k cells/ well and found 0.8-1.0 mm cell aggregate on 3rd day. My doubts are

1. is the cell concentration what i used is allowed or on what basis no of cells / well is decided.

2. is that a cell cluster or a spheroid or how to confirm that it is a spheroid.

3. it is looking so fragile. even while changing the media gently, the shape of spheroid is getting spheroid, so how to handle it.

Kindly help me in this regards
