i have been measuring zeta potential in both Deionized water and PBS (PH 7.4). The conductivity in case of deioinized water is zero and ioinic strength of PBS i have been using in very high which i believe screens charge on particles. Please suggest best way to measure zeta potential.
The ideal solvent (or rather liquid medium) is the one that the particles will be in for their application.
Zeta potential is a holistic interfacial property - it depends on the properties of the particle and the surrounding liquid.
So, please explain a bit more about your particles and their intended use.
I have drug loaded polymeric particles and their end application is for inflammation treatment
For intravenous use, X1 PBS buffer would be most appropriate because it resembles physiological conditions. For other routes of administration, use a medium that most resembles the environment the particles will encounter. In the case of oral administration, you might need to test multiple conditions, especially if the goal is for the particles to reach the intestines.
For media like X1 PBS, the main challenge will be measuring it. Commercial light scattering instruments struggle at such salt concentrations.
Note, that just because the charge is screen doesn't mean the particles will aggregate. It depends on the type of polymer.
