The Difference:
SYBR Green I (Cat.No.: HY-D1191, MCE) mainly binds to double-stranded DNA and produces fluorescence by binding to the minor groove region of all double-stranded DNA double helices. Its fluorescence is weak in the free state, and it is significantly enhanced after binding to DNA.
SYBR Green II (Cat.No.: HY-D1098, MCE) mainly binds to RNA and single-stranded DNA, and its fluorescence quantum yield for RNA binding is higher than that for DNA. And the fluorescence generated by its binding to RNA will not be quenched by urea or formaldehyde, so there is no need to wash out the denaturant before staining.
