I am working on QTL analysis and saturation of linkage map. my objective is saturation of linkage map and my question is that, what is the difference between construction of linkage map and saturation of linkage map.
Linkage map and saturation map/ chromosome mapping both the terms used by geneticists former determines what genes lead to which physical expressions in an organism's body, while the latter determines the physical location of a given gene on a chromosome's chain of genes.
Linkage map determines the gene order in a chromosome and can be done by recombination analysis. The physical location of genes is done by the cytological localization of DNA sequences (fragments) of any gene in a chromosome by in situ hybridization on (mitotic, meiotic or polytene) chromosomes.
In my understanding the construction of a linkage map is the calculation of a linkage map by the recombination frequency of molecular markers and phenotypic traits to get information which markers and traits are inherited within a population how often together.
And the saturation of a linkage map is just to recaculate the same map with a higher number of markers, especially for intresting areas in which pehotypic traits are located. By this you usually try to find markers that are linked more closely to the trait of interest. Therefore you get a better predictive power by the markers about the presence and absence of that trait.
In simple, Saturation of linkage map means increase of markers in density to cover each of the genomic reasons (Genome wide whole coverage of single chromosome or entire genomic chromosomes), While construction of linkage maps means assignment of markers having any numbers under study on given chromosome using various genetical software like Map maker, Genescan, cartographer, QTL ICImapping
