I have to perform an experiment "lysis profile of host bacteria after phage infection" in which i will check the reduction in bacterial growth at different MOIs (0.1, 1. 10) by taking O.D600 . After that i will check the surviving bacteria by plating them and calculating their cfu. What MOI should i adopt?
I have read various papers in which they have taken only one MOI (for example 0.1) . On what basis one MOI is selected for the experiment? Should i perform an experiment to calculate optimal MOI first? and then i should perform bacterial reduction assay?
I'm not sure what you mean by "lysis profile". Generally for doing a one step growth curve of a phage you would use an MOI of around 0.1 or less, the idea being that you want to minimize double infections. If you really want to see how efficiently you can lyse a culture, then it depends upon the time scale of your experiment. If you want one round of lysis only, then you need to infect all the cells with phage and hence would want to use an MOI of greater than 1. Usually to ensure infection of nearly all cells then an MOI of 10 is commonly used. On the other hand if you are going to infect and then culture for a few hours to allow multiple cycles of phage infection and lysis then there is much flexibility but 0.1 is a good value.
