John, even I don't have a reasonable answer for your question, most of the researchers that I know, would say that 4% would be the best fixation or at least far more common. I hope that helps
Hello.. as per my personal research experience 4% PFA is best, it provides best fixation for sectioning. The reason according to my opinion, may be that the time duration in between keeping the brain in PFA and sucrose solution should not be very long. So, if the time duration is short, then the 4% conc. of PFA will give enough fixation than 2%. If you want to use 2% PFA, then may be you should keep the brain in PFA (2%) for long period, even you will have to prepare the sucrose solution in PFA also, that is generally not required.
