I am evaluating ROS production by myeloid derived suppressor cells in terms of intensity of Dichloroflorescein by multicolor flowcytometry. i am also using FMO controls for the same. Presently i am interpreting data in terms of mean florescence intensity. i am little confused whether i should represent it in terms of mean or median florrescence intensity because using FMO for selecting exact gate for positive cells i am neglecting lower and middle range of positve cells in histogram which means i should use mean florescence intensity. i just want to know whether i am doing correctly or both ways are acceptable. i have also studied in various papers some have interpret it in form of mfi(sem) and some in median.
Both are acceptable, however, Median FI is more robust a measure vis-a-vis Mean FI
Hi Vinit, Median would always offer better alternative because it is cumulative distribution that you are representing with these variables. Since in a normal distribution both mean and median would be equal but this will be not be the case if fluorescence pattern is skewed.
