I'm growing HepG2 cells in 10% DMEM but they don't want to grow in a mono-layer. They grow vertically and in a pile. I get a ton of cells but I am not able to get a homogeneous suspension of them either for counting and plating.
Any advice would be really helpful. Thank you.

what do you use for resuspending the cells, how many rcf do you use and which flasks? Could you provide picture of the cells?
I used TPP surface treated flasks and for passaging acutase, very gentle pipetting (as HepG2 are more sensitive than HELAs) and gentle centrifugation 250 rcf and there was no problem in obtaining monolayer and resuspending the cells.
I would guess that the problem is your culture plastic which is not surface treated but maybe there will be another ideas.
