I am working with H9C2 cells at the moment, looking at hypertrophic effects of several compounds on the cells. I am using PCR for hypertrophic markers and total protein experiments, but would like to be able to measure cell size. So far I have tried immunostaining for alpha-actinin, but it is just not working in H9C2 cells at all, so it is time to move on to a new method! I have considered using H+E staining, but was told this is tissue specific, and would not work on cultured cells. What would be the best way to stain the cells?
