What solvent will be best for extract preparation and which will give good results for stigmasterol estimation? The tissue is part of a tomato plant.
Methanol will be the best for stigmasterol in case of UV and HPLC. I have performed this analysis with this compound.
Yes, Methanol is good for HPLC and UV. But for GC-MS you have to use DCM, chloroform, pentane, hexane etc.
Thanks a lot Sir for reply . But I have one doubt like it has been found that standard stigmasterol is getting dissolved only in Acetone and ethyl acetate. What will go wrong if I prepare plant extract in acetone.
In methanol standard is not getting dissolved completely.
MeCN/water i always use.
For hplc
Try sonicating the stock solution. It might dissolve.
With Methanol, use proportionsof Acetone/Eth Ac to solubilize the compound
N nope, Nothing goes wrong if u extract with Acetone/Eth ac, it rather increases the chance n even yield
Methanol or ethanol for UV and HPLC and Hexane for GCMS
@Raksha you can use acetone for UV but you have to use MeOH for HPLC. Use 70% MeOH or 70%ethanol and sonnicate at 40C. This may be help you out.
gudluck
:)
For best results on GCMS you would likely add your TMS derivatizing agent and an organic solvent like hexanes or n-decan.
For HPLC you can use also 1-Propanol. It works for sure. If you don't overload the column, you can obtain Gaussian peaks.
First check the solubility of standard if u are unable to do this tell me the name of standard
For GC/MS you can use n-hexane.
