Just use a concentration of mg/mL of BSA in water to have a full coverage of the citrate GNPs. You can speed up the reaction at pH 8 by adding 8 mL ethanol drop every minute when you stirred. An opalescent solution was observed indicating the formation of the nanoparticles. You need to add 8% glutaraldehyde solution for the stability of the particles. keep stirring overnight and then centrifuged at 15000 rpm for the nanoparticles to settle down. Remove the supernatant and you can lyophilize the pellets . After lyophilization, you can dissolved the particles in PBS buffer at pH 7.4 to prevent the denaturation of proteins.
