I'm trying to isolate some polyketides from Fungi and I need to know what is the best mobile phase that can be used for the clear separation and identification of Polyketides on TLC plates and also what is the best spraying reagent that can be used to reveal the spots of polyketides on a TLC plate?
TLC separation might be hard if you have a complicated mixture. You can try using EtOAc/Hexane or CH2Cl2/Acetone depending on the polarity of the polyketide. Most polyketides have conjugation systems so they can be easily seen at 254nm without using spraying solutions. Otherwise, you can use I2 (reveals the double bonds) and i think you will be fine
most polyketides have OH groups which can be deprotonated and make separation difficult. try multiple different solvent systems (with appropriate polarity) and add a little formic acid ~ 0.1-1% to your mobile phase. this may help give you better separation by keeping your compounds neutral.
