I am trying to get the best dilution which gives me 80% confluency next day for transfecting HeLa cells.
I supposed to get cell count 1.7X times the next day (which means if I am seeding 100,000 cells per well, next day I should see 170,000 no of cells in my well.
What is the best approach I can use to get best dilution for transfection?
Hi, what do you mean by dilution? Do you mean what should be the cell density for the day of transfection or you want to know the transfection protocol?

Dilutions means , I want to test at different cell numbers to see which one gives me 80% confluency next day for transfection. like 100,000 cells/well , 200,000 cells/well and so on till 600,000 cells/well for 6 well plate. I hope this clears the confusion.
Okay, I cannot give you exact numbers, but I usually seed 300,000 cells/well for 6 well plate format, and next day the confluency is 70-80% good enough for forward transfection. However, if you need exact 80%, I think you will have to calculate the doubling time of your cells and seed accordingly.
