what is the best method to know whether the cell undergo intrinsic or extrinsic pathway of apoptosis? is there any single test available to detect the exact apoptosis pathway?

caspase-8 is an initiator caspase for the extrinsic / death receptor pathway, casapase-9 is an initiator caspase for the intrinsic / mitochondrial pathway. You might check the expression of those upstream molecules.
Bid (BH3) / t-Bid as substrate of casp-8 is also a marker for the activation of the extrinsic pathway.

can we use flowcytometer to detect caspases 8, 9 in cells? can we directly use the serum or plasma cells to detect these??
