I am working on endophytes in cotton. I need to isolate endophytic fungi and bacteria from leaves, stems and roots. Plz suggest a protocol where we can easily exclude saprophytes and isolate only the endophytes.
surface sterilisation is the best way to eliminate saprophytes and "epiphytic" organisms from the plant part who's the object of isolation. We should make under consideration the hsitsological (specific tissues) and anatomical nature of the plant part.
Most scientific works on endophytic fungi and bacteria of crop plants was focused in roots, mainly for agrotechnical purposes.
