Generally during the transportation, DNA is often degraded, specially the High Molecular Weight DNA which is extremely fragile to external shocks. I am looking for suggestions/recommendations regarding export of High Molecular Weight DNA (more than 50-100 Kb) from one country to another, without losing the integrity of the DNA.
Does anyone have experienced this before?

Hello, the best and safe way is use of DNA vacuum concentrator or vacuum dryer under low vacuum. You can dry the samples in concentrator or under vacuum at 40*C and then you can send it under -20*С conditions without any problem. DNA will stabile for long time. I have experience good stability with 30-80Kb DNA samples.
I would suggest resuspending of the dried DNA over a period of 24 to 48 hours at 4* C. Don't try to rush this. Resuspension in either TE or Milli Q water.

Hello, Lyophilization is more aggressive for DNA than drying also more expensive technology.
Cycles of freezing and drying can be used shear DNA so I am a little doubtful if this is the best method. If 40 to 45 kb is enough, packaged cosmid clone DNA is completely stable. Native lysates in EDTA SDS buffer can be electrophoresed into very low procentage agarose gels using PFGE ( pulse field gel electrophoresis).
Lyophilization means you should freeze DNA first and then dry avoiding liquid fraction so it more aggressive for DNA than drying first and than freezing.
Finally !
Short-term storage (till 10days) at 4°C in Tris-EDTA.
Medium-term storage ( till 3 months) at –80°C in Tris-EDTA.
Long-term storage (years) at as –80°C as a precipitate under ethanol
Long-terms storage (decades) at –164°C or dried !!!! DRIED !!!
drop some drops of concentrated dna on whatman filter paper, let it dry in a sterile tube to avoid cotamination during transport
Just put it in the post and send it as a solution. Nothing it's likely to encounter under transport is comparable to the shearing caused by pipetting, which will normally shear DNA to about 40 Kb. About the only way I know to obtain DNA of higher molecular weight is to cast the cells into agarose before lysis and extraction, which is the method used in PFGE analyses. If that's what you've done simply send an agarose plug.

We have in the past spotted DNA samples on nitrocellose(or similar transfer medium of choice) allowed it to dry trimmed it and placed it in between Whatman paper. The sandwich was enclosed it in a plastic pouch and subsequently sent through the normal mail channels .
