the best way to preserve any fungal or bacterial species is to use 75% glycerol.. Take 1 ml spores of your fungi of interest and add about 500 microliter 75% glycerol and then keep at -80 degree C for a long time

I have saved fungal spp. in microbank. It has been successful for my lab and can be stored at -80 C for a long time.

for fungal species skimmed milk method, deionized distilled water and -80 C are some methods of storage.

Preservation of the pathogenic spore suspension at -80°C in glycerol is the best way as suggested by Dr. Latif.

Thanks Edward and Sosanka. well this is all about spore forming fungi. I have non spore forming fungi too. So any suggestion for those non spore fungi preservation?

I'm using a slant agar to preserve plant pathogens.....and every 2 weeks, I will sub-culture the fungi in a new PDA media...

A cheap and easy method is to grow fungi on squares of filter paper using PDA, once paper is colonized by the fungi, peel them and dry on a petri dish, when dry make small packages with alumin foil and paper bags and store at 4°C and -20°C. Other method is to grow fungi on PDA, then cut smal disc or squares, transfer to plastic vials and then add sterile destile water until they are completely cover. Store vial at room temperature.

Thanks Anam Moosa and Laith Al-Ani. But Both You did not mention any glycerol percentage to preserve fungi.

Storing culture slants under a layer of sterile mineral oil is one of the oldest, methods for long-term culture preservation. This widely used, for fungi that do not tolerate freeze drying.
