I am looking for an accurate method that is performed with the least equipment. Just insect whole body is available for me and haemolymph is not accessible because of storing insect in -80. Our lab equipment are HPLC, Spectrophotometer and micro plate reader. I already studied these papers: Shu et al.; 1997, Bergot et al.; 1981, Rembold and Lackner; 1985 and Gelman et al., 2007.
There are not many methods available for measuring JH that are easy or that don't require radioactivity or a billion tedious steps. See the following paper for one of the easiest and most robust methods based on HPLC coupled to a fluorescent detector.
