Which methodology is best and simplest for histological quantification of stained cells (ie count and measurement cell and tissue components of already stained tissue).
It could be on the microscope or through software. I just need the best and simplest way to get it done.
There is no such method. Softwares can help but the real thing is what you want to analyze. That is your first line...

Try Fiji that works on Imagej, thare are myriads of plugins for dirrefert tasks. I would suggest cellcounter for counting cells popultaions, analyze particles forautomatically counting elements and obtain euclidean morphological indexes, color segmentation plugin to quantify stains, color deconvolution for splitting histo images, fractal box counting for assessing non euclidean morphological patterns.
