Some pathogens require both antibody-mediated and cell-mediated immune responses to be neutralized and eliminated. A vaccine approach against these antigens must effitiently activate both components of the immune response.
In my case, we designed a recombinant vaccine against UTI. In this study, we took a sequence as adjutants in both humoral and cellular mediate immune response into consideration which we observed great result.

It mainly depends on what you use as antigen (peptide, whole protein, mRNA).
Usually a vaccine is given when there isn't an ongoin infection. However, there are some therapeutic vaccines (for example in cancer)
Usually, Th2 is more important for vaccines than Th1.
Ideally you could use a mixture of Aluminium salts and CpG/Poly I:C.
