I wonder if anyone has experience in freezing stem cells for further medical applications?
I am currently using 10% DMSO, 20% Autologous serum from the patients and 70% saline!
Saline makes no sense to me, as it adds to much water to the mix. Any suggestions? I generally use 90$ serum + 10% DMSO for PBMC and other cell banking.
The other question is related to thawing. I make sure to thaw quickly at 37 C and usually use compatible media (RPMI, McCoy, etc) supplemented with FBS and L-glutamine to wash the freezing mix and spin the cells. In medical labs, only Plasmo-Lyte which is a mixed salt solution is available. Does anyone know of any medical grade media or more nutrient medical grade solution?
