I am currently working on studying the anatomy of spinach roots, particularly focusing on the fine roots and the main tissues. However, I’ve encountered some challenges in identifying the best approach for embedding and sectioning.
Specifically, I have two main questions:
Would you recommend using paraffin or a resin such as LR-White or JB-4 for embedding? I am interested in achieving optimal preservation of the delicate structures while maintaining the ability to obtain high-quality sections for microscopy.Given the small size of the fine roots, what strategies or tips do you use to locate the roots within the block after embedding? This has been particularly challenging for me, as it’s easy to lose track of their position.I would greatly appreciate your insights or any protocols you’ve found successful for similar work.
