Hello everyone,
in your experience what would be the best cytotoxic agent to evaluate novel cytotoxicity/cellular viability assays? I am thinking of e.g. chemotherapeutic agents. Something that might give an overall nice concentration/dose-response curve, possibly low costs, low volatility, easy handling, and minor occupational hazard; kind of like a 1-agent quiver.

A positive control should be an established drug for that particular disease which you are working on. So, if you are working on cancer you should be using known chemotherapeutic drug. For instance, Tamoxifen could be a good positive control for breast cancer and Adriamycin for certain types of leukemia.

This does depend a little on the cancer type you are looking for. I would suggest either carboplatin or cisplatin as these are non-specific DNA intercalaters that interfere with DNA replication - you can titrate the specific concentration against your cell lines to establish the concentration-response curve. These are great as you can store them initially at room temperature and are cheap!

We have been using Cisplatin and Doxorubicin in cancer cells as a positive control in MTT assays. But it works better in some cancers line, you should use a drug that has a better effect on your cell type.
