I am interested in finding out what the appropriate dose of Gabapentin to administer to mice. Having read some papers- there is some information available for rats i.e. 30m/kg or 50 mg/kg IP, but not anything clear for the mouse. Has anyone administer gabapentin to mice? and if so what dose would you recommend to ensure analgesia without sedation?
I am a diabetic patient taking 2400 mg of Gabapentin daily. I weigh 220 lbs. If you break my weight down to ounces it comes to 3,520 ounces. Assuming my weight stays the same, dividing my weight in ounces by the mg per day (2400 mg), I have calculated it as 1.466 mg per ounce/per 24 hour period. Weigh your mice and dose accordingly. I would suggest 100 mg/kg IP or PO, but IP is probably better. Oral absorption of gabapentin in rodents (like people) is incomplete and decreases with high dosages. 100 mg/kg IP should not cause very pronounced behavioral depression or ataxia. Peak analgesic actions should be observed from about 30 to 120 min after injection. See attached articles.
I agree with Charles (above comment), you can go up to 100mg/kg po or ip, You get good efficacy in inflammatory models at this dose. I would suggest going po (less variable, at lease in my hands) and assess behavior 1hr post dosing
