for growing Agrobacterium that is I use 2-5 ml liquid in a 50 ml Erlenmeyer. That makes it even 10%. You want to grow it on a shaker or where? And for how long and at which speed?

Thank you very much for your answers. I really appreciate it. Actually, I want to grow bacterial strain in 3 liters of nutrient broth. For this purpose, I inoculated bacterial strain from nutrient agar slant to nutrient broth of 30 ml (10x3) for 24 hours.

So, I  was thinking that may be I can use 10 ml of bacterial culture (grown in nutrient broth) into 1 liter bottle containing 990 ml of sterile nutrient broth (1 % dilution) and incubate for 5 days with a constant shaking at 150 rpm.

In that bottle with 1 Liter of broth, there is still space equivalent to 200 ml and I was thinking that that space may be enough for bacterial oxygen requirement. Can you please clear me here?

The bottle that I am using is a 1 L reagent bottle and it has space even after putting 1 L of nutrient broth. Is it okay to use that bottle to grow bacteria in 1 Liter of nutrient broth or should i decrease the amount of broth to a lower value?
