I've been using ThermoFisher's chargeswitch plant gDNA kit for my daily DNA extractions, and I noticed that the wash buffer (W12) is not EtOH based (it does not require pre-adding any alcohol, nor does it smell anything alcohol, and it never completely dries up before the elution step - I guess it eventually will if you leave it long enough, but I usually dry the sample for 15-20min and I still see small water droplets in the tubes, which is nothing like using an EtOH based wash buffer). I understand that the way the kit works is through changing of pH (and thus the electric charge) to bind/concentrate the DNA to the magnetic beads and then release it back to the elution buffer after washing off unwanted impurities. But I'm just curious about how the washing actually works if the wash buffer is an aqueous solution.
