FLX pyrosequencing is used to perform DNA sequencing on a larger scale, compared to the original method, which could only manage short stretch sequencing.
FLX provides a high yield 1,000,000 sequences in a single 10-hour run with an average read length equal to 330 bp (Upto 1000bp) and It's highly accurate.
Now Focusing on FLX/Titanium:
In the Titanium technology loading beads are isolated from each other and there should therefore be no interference from adjacent beads. Read lengths are longer.
If you take the plain 454FLX, the primer sequences were
FLX A: 5’-GCC TCC CTC GCG CCA-3’ &
FLX B: 5’-GCC TTG CCA GCC CGC-3’
For the titanium FLX, the primer sequences were
Titanium A: 5’-CCA TCT CAT CCC TGC GTG TC-3’ &
Titanium B: 5’-CCT ATC CCC TGT GTG CCT TG-3’
The titanium kit also has the ligation library (Lib-L) and the amplification library (Lib-A), which are not included in the plain FLX.
