I am doing single spore isolation of Magnaporthe grisea using a serial dilution method but spores aren't germinating.
The protocol that I am using.
1. Flood the Petri dishes and slowly scrap the mycelium to release the spores.
2. Sieve out the mycelia using autoclaved cotton wool.
3. Carry out serial dilution using Autaclaved distilled water.
3. pippete a few mls of the solution and spread on PDA media
Kindly advise on what i can do to ensure they germinate

Growing M. grisea quite cumbersome. PDA maybe not enough. Have you tried Talbot's Complete Medium Agar (https://academic.oup.com/plcell/article/5/11/1575/5984612)?
