I am working on a heavy metal resistance study with bacteria collected from aquatic sites in an asbestos mine. Two of the metals being tested, NiSO4 and As2O5, lower the pH of R2A media below 6.00 and need to be buffered. Both NaOH and Na2CO3 buffers cause the metals to precipitate out of solution in liquid R2A. What could be used as a buffer that would prevent precipitation in both liquid and agar and still leave the metal available for intake by the bacteria?
