Hello, does anyone know what is a good dilution for a stock solution of alpha-glycyrrhetinic acid (alphaGA), the compound used as a chemical blocker of gap junctions on cells ? I have heard to is supposed to be ressuspended in DMSO, is that right ?
I plan to use it on a suspension of cells.
In most of the papers I have found, the final titration is written, but the precise nature of the vehicule is not clear...

Hello Richard,
when I used alphaGA for my experiments, the stock solution was prepared in DMSO at 100 mM and kept at -20°C. Then I diluted 1:1000 in culture medium to obtain a working solution at 100 uM.
The alpha GA does not dissolve well in solutions and you often see the powder in the bottom of the tube; so you need to:
sonicate the defrosted stock solution for 3-5 minutes,
dilute in medium and again sonicate (3-5min) the working solution before to apply on your cells.
Remember also to vortex well before and after sonication to help the dissolution of aphaGA.
