I mixed 20% Triton solution with liposome encapsulated with gold nano particle the UV Spectrum shows decrease (low) in the UV absorbance.
Untreated Triton 20% liposome (encapsulated with gold nanoparticles) shows very high UV absorbance. Why the difference in UV absorbance intensity. Whats going on the sample.

Why did you use Triton X 100 here? To disintegrate liposomes or to stabilize gold nanoparticles? Liposomal disintegration could result in the formation of large micelles with Triton X 100. May be the change in absorbance (I presume its golds absorbance you are talking about) you see here because of this phenomena.
Hi Anish, I used Triton to rupture the complete liposome. It's not for stabilizing the gold nanoparticles.Thanks for your reply.
You may need to evaluate any solvent dilution happens while treating liposome with 20% TritonX100
I have experienced the same issues. You may try heating your sample ~ 60 degree for 2 min and then keep at RT for another 15 min. You may also sonicate briefly and then try again. Be sure to take the same percentage of Triton-X solution as a blank (it has very high uv abs at 290). I would suggest to try with 1% SDS. In my case that worked far better than Triton-X. Good luck!
