I am conducting an experiment using a 1.5% agarose gel with ethidium bromide as the nucleic acid staining dye. In my experiment, a minimum of 850 ng of DNA was required to visualize the DNA band. However, ethidium bromide is known to detect as little as 10 ng of DNA. Could anyone provide insight into why this discrepancy occurred?
