What does the alpha level indicate in RSM? Why are conc. and time negative in my test combination??

Why is the alpha value negative in my RSM design? As far as I know, I can choose the range of low and high values independently. When I'm doing so, the alpha value becomes negative, and this value comes as a test combination as well. Am I doing something wrong or missing something? Can anyone please help me out?
