Hydrolysis of amino acids should be out under air (oxygen) free conditions for best results. There are specific hydrolysis tubes available that can be evacuated and purged with nitrogen (oxygen free) before heating, but any suitable sealed tube that can be purged before use should suffice.

Could you recommend a suitable brand for sealed tubes to carry out hydrolysis? On the other hand, how could I adequate any tube to perform hydrolysis under nitrogen atmosphere? I have a graphite digester that can reach temperatures up 600°C. At the same time, it can hold tubes without cover of 400 ml.

Thermo fisher supply a range that are suitable but there are plenty of other manufacturers about. Here is a thermo catalogue n/o ( 29570). You can see a picture of the tube so you may have something similar already you can use.

Thank you, I think it is necessary counting on this equipment for an HPLC process line. I mean, until today I have read that vaccum must be applied to most aminoacids, and a nitrogen atmosphere for triptophan analysis. Could you tell me if this equipment (from Thermo Fisher or others) and components are essential and more efficient for this process line? Because this is a part of the implementation in my laboratory.

We use the vacuum tubes as they produce relatively quick and reproducible results with small amouts of sample, although evacuation of the tube must be done carefully to avoid sample loss . There are many ways carry out hydroylsis, enzymes for example, so you must decide what your key requirements are before making any purchases.
