I am trying to prepare water in oil vaccine but I found that everytime the mixture was not homogeneous after overnight keeping at 37C or 4C. What are the faults that I can avoid. Also, how I can sure that the mixture is mixed well.
Thank for your response Dr. Hanif but I need your experience in the type of homogonizer (magnetic stirr or blinder... ) and average speed should be to get good results.
I neither used blinder nor magnetic stirrer for oil based vaccine homogenization it will not work for absolute homogenization, I will recommend to use a Homogenizer specific for vaccine homogenization.
I would suggest to use device with fastest and strongest homogenization effect. I used ultraturax, but if you do not have such or similar device, try with some powerful blender rod, not magnetic stirrer. Adding emulsifier is also an option. If the emulsion is properly done and the inactivated vaccine stratifies overnight, strong shaking prior to use will restore the emulsion.
