I am looking for a bit of advice about solubility of fluorescent lipids in oil (DEP or mineral oil).
my protocol to create an oil+lipids mixture says (at least) 5 cycles of heating 10min at 80C and 10 min of sonication.
after that, the mixture is perfectly homogeneous...BUT you will not find any slightest trace of fluorescence coming from the fluorophore.
The worst result comes from DOPE PEG(2000)-N-Cy5; but also NBD-EggPE gives problems and so on other fluorophores.
(I need to work with at least two well-separated colors. )
if I add it to the final lipid-oil mixture only with vortexing, it will not mix at all, and - on top - chloroform will not evaporate and its remints are damaging for the droplets/vesicle creation.
What shall I do?
