Mutation in restriction site, try other enzymes

PCR is very sensitive so you could be amplifying un-ligited excess insert.

Depending on your original restriction enzymes, you might not have the same restriction sites present anymore.  Try another enzyme that cuts in areas that "flank" your insert.  Or you could find an enzyme that will cut in both your vector and your insert to give a unique digest pattern.
