I have been trying to optimize the biofilm formation for enterobacter cloacae for which the biofilm formation is sparsely studied. i have been facing great difficulty in the C.V assy. I am getting the ring formation in the control well tube . why is it so ?
there is no contamination in the inoculated well/ tube . kindly suggest

We have had experience with E. cloacae for biofilm formation on jug filter respect to P. aeruginosa. E. cloacae did not produce biofilm.

..unicolated is such a non scientific word - please multiple inoculated media. what was the control? you only mentioned E. cloacae? what other bacteria was present in the inocculum? What type of tube did you use?
please elaborate on the question and I will help you I have studied many biofilms on different surfaces. one of the reasons is if you word with multiple cultures is the difference in generation times. Please send more info
If you study biofilm production it must be for certain purpose. Why not onto a surface of medical or industrial importance. Why do you not use a sterile latex tube or titanium disc or surgical steel or what was the purpose of the study? Then you can also use more specific techniques like SEM of Confocal microscopy.
1. You need to keep the setup in an environment that the temperature is up to 38-40 degrees Celsius, to prevent aerosols contamination. 2. The laboratory where the experiment is being carried out should be air sterilized possibly with UV in a controlled experimental cupboard.

Sir, actually there is no study on enterobacter cloacae SBP-8 (sparsely studied) so I want to know the biofilm formation under various physiological conditions . Further I would like to study about the correlation of curli with biofilm formation.
as a positive control I have used salmonella ,control is media without inoculum.......... kindly suggest as I am getting absurd readings
