What could be the control while performing the Mito Stress experiment using nanoparticle as a drug in XFe96 analyzer?

I want to measure the effect of Nanoparticles on macrophage metabolism, but I'm confused about the control. How can I be sure the result I'm getting is due to the effect of nanoparticles on cells or the effect of nanoparticles with the measuring system? Any suggestion please.

Also add them to media containing wells only?! be shure to have multiple wells as controls as well as your active measurment - the seahorse is not very exact.
