Accumulating research shows that NF-kB is a critical transcription factor in many diseases such as tumors, RA, psoriasis,etc. It could control expression level of cytokine including IL-6, IL-8. In some study of treatment of autoimmune disease, therapeutic agents could down-regulate the mRNA and protein level of NF-kB, thus blocking the process of disease. NF-kB signaling is a complex network which contains a variety of signal proteins, I wonder which signal protein could control the transcriptional level of NF-kB?
NF-kB activation can be induced by variety of signals, including CD40 mediated signaling and TLR4 mediated signaling, for example in B cells. One way of measuring NF-KB activation is the detection of nuclear translocation of NF-KB p65. Activity itself can be directly measured by chemiluminescent NF-KB o65 transcription factor assay (Thermo). Raw-Blue cell line (InvivoGen) can be used for measurement of NF-KB activation. Cells were stimulated and the subsequent production of SEAP was detected by Quanti-Blue (invivoGen). Other than that, any immunology text book will provide you answers. Good luck.
