Recently, I have performed the overactomy surgery. I am interested to give the treatment of β-Estradiol + progesterone as an intraperitoneal injection. Litrature suggest the normal 5–45 pg/ml concentration of estrogen in serum. But I want to know how to calculate the concentration of these steroids as per the body mass, for example if the body weight of individual rat is around 120 gram.
We used estradiol and progesterone in our research with cattle. The concentration will depend on the type of hormone used (free or conjugate), the type of diluent (oil-based or alcohol-based or water-based). The site of injection will affect the levels (subcutaneous, intramuscular, intraperitonal, etc).. We typically used estradiol benzoate to get a more predictable response. We used free progesterone. One can use very small silastic tubing and pack the tubing with powdered estradiol and progesterone and insert these beneath the skin (seal ends with sliastic glue).
