I'm planning on doing my first experiment with THP-1 derived macrophages by inducing differentiation with PMA. I've read that PMA can also induce apoptosis in THP-1 cells and was wondering how much cell death I can expect?
The literature suggests that concentrations ranging from 100ng/mL down to 5ng/mL PMA are sufficient for THP-1 differentiation.
In the paper attached below, the study shows that in the Trypan Blue dye exclusion assay, the highest PMA concentration tested (1000ng/mL for 72 h) resulted in 83.5% THP-1 cell viability while 100ng/mL PMA-treated THP-1 cells at 72 h resulted in ~92.55% cell viability.
The study concludes that exposing THP-1 cells to 15ng/mL PMA for 72 h is sufficient for differentiation with a higher cell viability.
