I had to restart the cryostat last week. Everything is working well except the object temperature which is block at zero degrees. (Chamber temperature is at -22°C) Have you any tips ?
I am probably not understanding your question well. Every item for cutting has to be at -15 to -30C (varies slightly between different tissues sometimes). If some onetemperature is high, leave in chamber for sometime till the temperature is equilibrated -22C or so, If you want to cut tissue at 0C, will not work.
Than problem in cryostat. Need to call technician or use in another lab. Only you can do temporarily, you can dump dry ice inside, but be careful, it is CO2, do not leave your face longer time in the chamber. I have used one in Stanford 2 years ago, throwing dry ice in the chamber, it did work, that is temporary solution. I will not recommend permanent solution.
Subhash is correct. There is a problem with the cryostat cooling that needs to be attended to by a repair person. Basically, it's like a freezer that is not working properly.
