We recently aquired a fluorescent PNA (Peptide Nucleic Acid) probe, which is lyophilized. This probe is to be used in FISH essays. I was wondering what is the best way/buffer/solvent to resuspend said probe. I've been told by some that formamide solutions are best for the FISH protocols, but others ources recommend nuclease-free water or some TE buffers. I was wondering which is the best way to solubilize the oligos.

Keep in mid that:

  • These oligos are not made of DNA//RNA. The oligonucleotide has the sugar backbone replaced by N-(2-aminoethyl)-glycine units.
  • The probe is labeled with TexasRed fluorophore.
