We recently aquired a fluorescent PNA (Peptide Nucleic Acid) probe, which is lyophilized. This probe is to be used in FISH essays. I was wondering what is the best way/buffer/solvent to resuspend said probe. I've been told by some that formamide solutions are best for the FISH protocols, but others ources recommend nuclease-free water or some TE buffers. I was wondering which is the best way to solubilize the oligos.
Keep in mid that:
- These oligos are not made of DNA//RNA. The oligonucleotide has the sugar backbone replaced by N-(2-aminoethyl)-glycine units.
- The probe is labeled with TexasRed fluorophore.

70% (v/v) formamide (deionized)
0.25% (w/v) Blocking Reagent (see recipe below*).
10 mM Tris·Cl, pH 7.5
Store up to 6 months at -20
* Blocking solution - maleic acid buffer (100 mM maleic acid, 150 mM NaCl, Adjust pH to 7.5 with NaOH).
You can store up to 1 year at 4°C.
