I am going to treat animals with the drug Astemizole. I'm looking for a suitable vehicle and concentration that can dissolve the drug without being toxic to the mice. Anyone who can help me?
Astemizole is soluble in water. So, if you are going with oral route in mice, you can dissolve it in water and can dose the animals. Solubility of this compound seems to be grater than 400 mg/Liter or 400 ug/mL. This should be enough for your work. Secondly, for IP or IV route, I would go with normal saline. Hope this helps
If you want to give an oral administration, you can use tween 80 and CMC suspension.
Weigh the compound according to the dose you want to administer. Transfer the compound into motor and add 1 or 2 drops of tween 80 and grind it to an even paste and start adding 0.5% Sodium carboy methyl cellulose (sodium CMC) solution and keep mixing the compound slowly and make up the suspension to the desired volume to make a stock concentration. From here the compound can be further diluted basing on your requirement. in sodium CMC. You can sonicate the stock solution for 5 - 10 minutes to get even particular size.
I think bio-availability depends on molecule and not on this formulation. As I have mentioned about the formulation, tween 80 gives a negative charge to the molecules, due to which they repel from one another. Secondly, this facilitates the molecules to be suspended in CMC. But, when you are doing the animals it is recommended to vortex the suspension for each dose. I forgot to mention about the intravenous dosing. You can use 20% HPbCD (Hydroxy propyl beta cyclodextrin). You can solubilize your compound in DMSO and add cyclodextrin solution to it and make a clear solution. I think final conc. of DMSO shouldnot exceed a specific percentage (may be 3-5 %).
