Certain sprays or coatings are needed to confirm the presence of lutein in marigold oleoresin while working with TLC. Can anybody suggest such sprays (with atomizer) and how to prepare them.
You can use a solution of cobalt Chloride: issolve 10 g of CoCl2 in 250 ml of distilled water, then add 50 ml of sulphuric acid and gage to 500ml. Spray this solution over your TLC and heat at 100 C for 5-10 minutes. The compounds should appear as violet or pink spots.
Also, you can use sublimated iodum: put your TLC into a sealed chamber saturated with sublimated iodum.
Hmmm....so Dr. Olga you can suggest that no sprays are needed for identification.....if then what sovlent system can be followed for obtaining the best separation of the compounds from the marigold oleoresin extracted with Petroleum ether. And what is the probable Rf value for it?
Dear Surendra, I mean you can see the lutein because is a colored spot, may be it appears others minor spots but lutein is the major component. Of course you can use some sprays as other researchers advice you.
