I have performed a tissue homogenization/extraction protocol using methanol and chloroform as the solvents. The aliquot being using for BCA analysis contains both solvents. I understand that the same diluent must be used for all samples and standards. I have unsuccessfully used HPLC grade water. The sample does not mix well enough with the water to confidently make serial dilutions. Does anyone have recommendations? I am looking for a way to either improve the mixing of the methanol/chloroform samples with the water through possibly a suitable detergent or emulsifying agent; or another diluent other than water that would mix with the methanol/chloroform sample.
