I have collected several wild mushrooms during the past few years and kept some of the interesting mushroom species tissues in CTAB vials and stored in deep freezer. So I want to know the viability of those tissue whether the chances are good or poor to get DNA from those tissues.
1. if you are targetting for DNA isolation, the chances of succeeding will be fair with the recently stored cultures rather than the very old ones.
2. Selection of extraction protocols adds up for the quality and quantitative results
3. Degradation of DNA during the extraction protocol is one of the issues with respect to wild mushrooms, which needs to be taken care.

Deep freezer is a very general term, you need to tell exactly the temperature at which you have stored the fungi. You may get good yields at -80, however temperature higher than this (-20) will surely affect the yield and and most important the quality of DNA.

The fungal tissues has been kept in home fridge only...! So, but, these all been kept in a particular solution which is normally use to keep safe the fungal tissue for long time.

My bet is that you can isolate DNA that would be good enough for PCR if you use a CTAB isolation procedure. Do not try a silica matrix-based kit without prior chloroform purification.
