According to the paper of Paul Ridker and colleagues (Circulation. 2000; 101:1767-72), they used stored plasma obtained at baseline from each control subject. Plasma was thawed and assayed for IL-6 by use of a commercially available ELISA (R&D Systems). Median control men plasma concentrations of IL-6 at baseline were as followed (all in pg/mL plasma; P=0.002 to 0.006): All participants – 1.46; Non smokers – 1.46; No hypertension -1.40; No hyperlipidemia – 1.41; No diabetes – 1.42; No obesity – 1.38; No family history of CAD – 1.48.

So, the above (and several other papers) all show average IL-6 plasma concentration at least 10 times lower than that indicated in your question. Please adjust your data.

Hi Chloe,

It depends on the context of when the plasma was taken and who it was taken from. 

Systemic IL-6 can be higher during lots of different types of illness or inflammatory event but as the above poster suggested, 15pg/ml would seem to be an error in healthy rested subjects.

Are all the values high or just some?

What was your measurement system? ELISA, bead array etc? If so what company etc?

A value that high may be due to some non specific binding in your assay.

It depends. In this study: http://www.ncbi.nlm.nih.gov/pmc/articles/PMC3958530/pdf/pone.0092319.pdf I analyzed plasma with Cytometric Bead Array in a Flow Cytometer. Healthy subjects had between 0 and 3,5 pg IL-6/ml, the Hematology patients between 0 and above 1000 pg/ml, with a median of 330 pg/ml.

I have also analyzed plasma from patient with different infections. There, the median was around 500 pg/ml.

IL-6 serum levels vary a lot and I don't think levels in normal individuals in two studies would match. The mean IL-6 levels in sera of healthy normal individuals that we recently assessed were 5.9+1.01 pg/ml (range 5-7 pg/ml).

basal levels of IL-6 in plasma samples varies with geographies, ethiologies, health status.

In healthy subjects in Indian population it ranges from 0.5-5 pg/ml

In eosinophilic subjects it ranges from 3-18pg/ml in plasma

does anybody know what the normal blood plasma level of IL-6 in mice?

I think that it depends in many factors but it can be several pg/ml in dependence of used method and manufacturer
