We want to take AFM image of quantum dot -DNA array by adsorption of sample on APS mica but we could not get clear topography of quantum dot. Please give me suggestions.
Please clarify further. Have you been successful to get atomic resolution images of the hexagonal lattice on untreated mica? Have you been successful to get high-quality images from the AP-mica? Are you certain that your QD-DNA array is absorbed? Are you certain that it is firmly anchored? What exactly do you mean by "clear topography"? Are you aware of whether you are convoluting the radius of the QD with the radius of the tip you are using, so you perhaps will not "see" the smaller particle?
Otherwise, I suggest the analysis will be a matter of trail-and-error, starting from a known, comparable surface to the unknown. The gain settings might for example be comparable between the AP-mica and the QD-DNA/AP-mica. You may want to also consider how firmly you are contacting the surface during the scans.
I can help you out : But need more details : Are you trying to get it done in ambient conditions ? What are the sizes of QD's ? Have you characterized your junction ( Tip and the sample ?) May this should be done before you actually do the experiment. These test will give you what is the conditions under which you are doing the measurements ? ( in terms of stability, noise etc).
